Gain is how sensitive the gyro is to yaw movement.

The TB/Align gyro will work either way. I would use the radio if I had a choice

With the yellow lead not connected to the RX the gyro is in the HH mode only and gain is adj by the pot on the gyro. When the yellow lead is connected to the Rx (CH 5?) then the TX controls the gain and whether the gyro is in HH or Rate mode. Check other posts for the TX setup (swithcable gains) that match your TX.
