I would suggest overnight at the very least, 24 hours is better.
The instructions say to apply the bead, re-assemble, but do not tighten down all the way, stop when it starts oozing out between the parts, then wait at least two hours better is overnight. Other guys who use it just go ahead and start using their engine after they re-assemble everything.
