From all the information you have provided, I have come up with a general fix.
1. Plug in the antenna.
2. Make all power and data connections according to the manual.
3. Read the manual, and make sure your software is setting all the parameters correctly.
4. Plug in the batteries
If the above fails, contact the manufacturer, or try giving us more information so we can be less sarcastic in our answers. ;)

In addition to the above…
Correct antenna choice and the selecting 9600 over-air data rate (as opposed to the data rate at the serial port) is critical for long range. You’ll need a high gain directional antenna on both sides to realize the range listed in the literature. Since that is impractical for the aircraft, you’re not going to get that 40mile range.
As Radiohound said, we’d love to help but don’t have enough to go on…

Another thing worth adding: you need a nice clean power supply and a quiet RF environment for radio receivers to work well at long range. These are hard things to achieve on a plane and it takes some expertise to reduce these enough to get really good range.
I suggest becoming an amateur radio operator with an advanced license of some type as a good step to understanding the issues involved.
